These results in short-term cell culture systems do not appear to translate into an effect on IgE synthesis in humans in vivo. As shown in Figure 2, total serum levels of IgE (comprising IgE-omalizumab complexes) increase and then plateau at steady-state omalizumab. If there was a reduction in newly synthesized IgE, the levels of total serum IgE would be expected to subside over time. This is not the case. In addition, serum levels of free IgE tend to increase with waning omalizumab concentrations towards the end of each interval (2-4 weeks) between doses of omalizumab.
