If we assume a common mechanism for the topoisomerase reactions illustrated in Figs. 2 and 3, then this mechanism must possess certain features. DNA binding and cleavage are clearly required and the positions of broken ends relative to another single- or double-stranded segment of DNA must somehow be altered before rejoining. As the product DNA is covalently closed, this break can only be transient and the enzyme must be capable of reforming the broken bond(s). Phosphodiester... [Pg.77]

Bleomycin, phleomycin, tallysomycin, and chartreusin are antibiotics which effect cleavage of DNA strands, and gel sequencing methods have been used to determine their preferred sites of binding and cleavage. The products... [Pg.208]

The preceding FID analysis of several metallobleomycins verified that the DNA binding site-selectivities of Co(III) and Fe(III) bleomycin parallel each other quite closely and also correlate to the established cleavage site-selectivity of Fe(II) bleomycin + O2. Notably different, Cu(II) bleomycin did not exhibit an FID assay response under conditions used for the other metallobleomycins. This outcome was likely due to the weaker DNA binding and fast exchange of Cu(II) bleomycin relative to other metallobleomycins (48) and was no doubt exacerbated by the presence of a competitive binding fluorophore. In spite of this limitation, the FID assay is likely to contribute to the study of additional metal complex-DNA interactions in the future. [Pg.73]

Abstract Several artificial anthraquinone-carbohydrate hybrids were designed and synthesized. A selected niunber of hybrids did bind to DNA via intercalation in a sequence-selective manner, leading to inhibition of DNA cleavage by DNase 1, whereas other hybrids cleaved DNA with base selectivity upon photoirradiation. This hybrid system was found to be very important for both DNA binding and photocleaving. [Pg.285]

The amino acid 58 was used in the solid-phase synthesis of sequence-specific DNA binding polyamides containing N-methylimidazole and N-methylpyrrole amino acids <96JACS6141> and it was also reported that the imidazole-acridine conjugate 59 could effectively catalyze the cleavage of t-RNA <96TL4417>. [Pg.157]
