Diethyl mercaptoacetal has been prepared by treating diethyl bromoacetal with potassium hydrosulfide 3 by the reduction of 1,1,11 -tetraethoxyethyl disulfide 3 with lithium aluminum hydride by reduction of 1,1-diethoxyethyl benzyl sulfide,3 41,1,1/,1/-tetraethoxyethyl disulfide,3 and 1,1,1, 1 -tetracthoxyethyl polysulfide 3 with sodium and liquid ammonia. The method described is adapted from the last-named preparation. Dimethyl mercaptoacetal has been prepared by the same methods.3...

DMS is formed during the biodegradation of organic sulfur compounds and by the biological methylation of sulfide and methanethiol. Precursors of DMS are methionine, S-methylmethionine, dimethylsulfoniopropionate (DMSP), dimethylsulfonioacetate (DMSA), dimethyl sulfoxide (DMSO), methionine sulfoxide and sulfone, trimethylsulfonium salts, S-methylcysteine, homocysteine, dimethyl disulfide (DMDS), 2-keto-4-methiolbutyrate, and 2-mercaptoacetate (thioglycollate). 

A straightforward synthesis of this system involves the addition of mercaptoacetic acid or its ester across the double bond of the azomethine linkage. 3,3-Dimethyl-2-ethoxycarbonyl-l-indolene (245) reacts with appropriate mercaptocaids (or esters) to give the corresponding thiazolo[3,2- ]indol-3(2//)-ones (246) (74LA206) (Scheme 60).

Two different routes to 5,7-dimethoxy-6,9-dimethyl-l,2-dihydro-3H-pyrrolo[l,2-a]indol-3-one (137) were developed by Vice and co-workers [48, 49]. The first route (Scheme 19) was based on addition of a methylthio group at C-3 of 5,7-dimethoxy-2,3,6-trimethylindole (134) using methylsulfenyl chloride [48]. Treatment of the product (135) with lithium diethylamide and methyl iodoacetate gave ester 136. Reduction with mercaptoacetic acid followed by base-catalyzed cyclization then gave 137.

The reaction of 8-aza-5,7-dimethyl-2-(trifluoromethyl)chromone (133a) with alkyl mercaptoacetates afforded bicycles 181a,b. When the reaction time and the amount of EtsN were increased, acyclic derivatives 182a,b were isolated [91]. A similar reaction of pyranopyrazole 135 proceeds at the C-6 atom followed by pyrone ring opening and intramolecular condensation of the aldol type to give compound 183, from which heterofused coumarin 184 was obtained [59] (Scheme 59).
