A method that has been widely used for structural studies of polysaccharides has been that of exhaustive methylation, followed by depolymerization and estimation of the various methylated monosaccharides. Kircher (190) assessed the technique by applying it to polysaccharides that had been previohsly studied, such as guar gum, starch and dextran, and showed that the results obtained by GLC were in good agreement with those obtained by other methods. The procedure has, since then, been applied to a number of polysaccharides of unknown structure. Methylated sugars have also been examined (IS6) in a GLC-mass spectrometry system. This technique will become increasingly important for the confirmation or identification of GLC peaks in methylation and other studies. [Pg.316]
