Dendrimers convergent approach

A dendrimer-based approach for the design of globular protein mimics using glutamic (Glu) and aspartic (Asp) acids as building blocks has been developed [151]. The preassembled Glu/Asp dendrones were attached to a 1,3-bifunctional adamantyl based on a convergent dendrimer synthesis strategy (see Fig. 28). 

Frdchet et al. reported the synthesis of a unimolecular micelle (Fig. 6) generated by a convergent approach starting with 3,5-dihydroxybenzyl alcohol, as the key branched monomer unit [65]. There is a plethora of examples representing dendrimers with metal-binding sites on the periphery. Polycarboxylates were the simplest, early examples [179], although dendrimers possessing more complicated ligand complexing centers have invariably been built via a convergent approach. 

The convergent approach to dendrimer synthesis has evolved considerably over the past dozen years. With the benefit of hindsight, it is clear that the key criteria for a successful convergent synthesis include ... 

The convergent growth approach to dendrimers [1] first introduced [2] in 1989 at the IUPAC meeting on macromolecules in Seoul, Korea, has provided a useful alternative to the divergent methods exemplified by the work of Tomalia et al. on PAMAM dendrimers [3] and Meijer et al. on polypropylene imine) dendrimers [4], Today several hundred papers have exploited the convergent approach to dendrimers to prepare a variety of synthetic functional macromolecules of unparalleled structural precision. 

The first report of all-phenylene dendrimers was presented in 1990 by Miller and Neenan [10], Via a convergent approach, they coupled arylboronic acids to arylbromides under Suzuki-type conditions to synthesize the first two generations of polyphenylene dendrimers based on 1,3,5-triphenylene building units (Scheme 1). 

Alternatively, dendrimers with chirality in the molecular periphery can also be constructed by the convergent approach starting from chiral monomeric units. 

Instead of a single carbohydrate moiety, a small cluster can be attached to each terminal functional group of the dendrimer 36 (Fig. 22),86a or used in a convergent approach wherein the cluster is attached to branching units, which in turn are finally linked to a suitable core.83a,87a,b...
