Darvon - Propoxyphene hydrochloride

Eight of the nine patients experienced headache. Onset ranged from 4 to 36 hours, and the mean time of onset was 13.3 10 hours. Duration ranged from 8 to 60 hours, with a mean duration of 30.6 19 hours. The headaches ranged from severe to mild, but usually were mild by the second hospital day. Five individuals headache responded to Darvon (propoxyphene hydrochloride, manufactured by Eli Lilly and Company, Indianapolis, Indiana) or codeine.

Darvon , d-propoxyphene hydrochloride, is a synthetic, nonantipyritic, orally effective analgesic, with similar pharmacological activity and effects to codeine. Darvon is not a narcotic but can be substituted for codeine, and is useful in any condition associated with pain. Chemically, this analgesic is not analogous to codeine or to morphine. 

Propoxyphene Hydrochloride, USP. Propoxyphene hydrochloride, (25.3/f)-( + )-4(dimethylamino)-3-niethyI-l.2-diphenyl-2-butanol propanoate hydrochloride (Darvon. Dolene. Doxaphcnc). was introduced into therapy in 1957. It may be prepared by the method of Pohland and Sullivan. It occurs as a bitter, white crystalline powder that is freely. soluble in water, soluble in alcohol, chloroform, and acetone, but practically insoluble in benzene and ether. It is the a-C +) isomer the a-f-) is-omer and P diastcreoisoiners are far less potent in analgesic activity. The a-f-) isomer, /evo-propoxyphene. is an effective antitus.sive (.see below).

Another example is propoxyphene [33]. The (2R,3S)-enantiomer, sold as the hydrochloride and once marketed as Darvon , is an analgesic. The 2S,3R enantiomer is an antitussive and was once marketed as Novrad . Today, neither is marketed in the United States. Darvon was removed in 2010 and Novrad removed about thirty years earlier. The scientists at Lilly who discovered these compounds were clever in their naming. NOVRAD is the mirror image of the name, DARVON.
