Cultures with Low Mutant Background

The above technique works equally well for cultures of either phenotype. In addition to this dilution procedure, TK - cultures can be maintained free of revertants by culturing them in Fiop supplemented with BUdR (50 pg/ml). Two days prior to use, the cells are centrifuged and resuspended in fresh Fiop to rid the culture of residual BUdR.
