Conformation of sugars

Direct experimental evidence for the existence of an ordered conformation of sugar nucleotides in solutions has been reported by Hirano,344 who observed characteristic optical-rotatory changes for a series of these compounds upon transition from water to concentrated urea solutions. The structural requirements for such an ordered conformation are still not clear. However, data at this point, based on indirect kinetic evidence from hydrogenation and hydroxylamin-olysis reactions (see Section IV, p. 360), seem to accord with the hypothetical model just described. Further studies on the conformations of sugar nucleotides in solution are highly desirable. 

NMR measurements to deduce conformations of sugar rings, three-dimensional structures, and the degree of conformational flexibility in various parts of N-linked oligosaccharides (Fig. 4-21). Measurement of C-O-C -C spin-coulping constants is also of value.270 Use of multidimensional NMR has permitted analysis of mixtures of cellulose oligosaccharides.269... 

Hudson s rules of isorotation are used for the determination of the configuration at the anomeric centers in oligo- and poly-saccharides and Korytnyk has discussed some extensions of these rules. Reeves has demonstrated that studies of the optical rotation in cuprammonia solution can give valuable information on the types of linkages and also on the conformation of sugar residues in a polysaccharide. Bourne and coworkers have shown that some o- and /S-D-glycosidic linkages display characteristic absorptions in the infrared at 844 and 891 cm, respectively.
