Chromatographic studies purification process

In general phenolic lipids have been separated from natural sources for compositional studies and structural determination by solvent extraction and chromatographic techniques. The individual component phenols of the major phenolic lipid (CNSL) from Anacardium occidentale have assumed some significance in certain chemical applications and detailed purification processes... 

Blanche et al. [45] showed that the P-CAC technology is very promising for the purification of Plasmid DNA at preparative scale especially when resins with low binding capacities for the product of interest are used. The aim of the study was to purify the Plasmid DNA out of a clear lysate of E. coli. The lysate containing RNA, nicked DNA, as well as the Plasmid DNA was loaded onto the annular column filled with Poros 20 R2 beads as the stationary phase. The chromatographic process for the purification is shown in Fig. 7. 

Arindam Bose (Pfizer Central Research) further discussed the ICH documents and presented a rationale for the recommended combination of test procedures and process clearance validations required to demonstrate that marketed biopharmaceuticals are free of adventitious agents. He showed that testing of Pre-Seed Stock (PSS), the Master Cell Bank (MCB), and the Working Cell Bank (WCB) is required to demonstrate that they are free from contamination by mycoplasma, bacteria, molds, and yeasts. In addition, viral clearance validation studies must be performed on scaled down versions of each chromatographic step and the viral inactivation/removal step employed in the product purification scheme. Finally, clearance studies must be conducted with a panel of relevant and model viruses (typically three to four) to establish that the purification scheme will indeed purge any viruses that may be inadvertently introduced during processing. 

F]fluoro-2 -deoxyuridine, [18F]FdUrd, 334, used in tumor chemotherapy and detection of tumors in human brain and lung, is produced for routine clinical use in a microcomputer-controlled process consisting of addition of 18F2 to 3, 5 -di-0-acetyl-2 -deoxyuridine 335, hydrolysis and elimination of AcOH from the 18F-triacetate 336 and chromatographic purification of [18F]FdUrd in tandem columns of ion retardation resin and alumina (equation 143)322. (The mechanism of elimation of AcOH from the 18F-adduct 336 has not been studied.)... 

The biosynthesis, occurrence, and biological activities of piperidine alkaloids arising from lysine have been studied Punica granatum. Piper species, and Lobelia inflata). The isolation process, chromatographic purification/analysis, and structure elucidation of the significant alkaloids are discussed.
