Anticholinesterase blockade of the ACh hydrolytic capacity of AChE induces a short-term hypercholinergic activation in the brain, leading by a rapid yet long-term feedback process to accumulation of an excess of AChE-R and modification of the cholinergic status in a manner affecting BBB properties at a later phase.

Contraindications Glaucoma, obstructive uropathy, obstructive disease of Gl-anti-cholinergic tract, paralytic ileus, intestinal atony of the elderly or debilitated patient, unstable cardiovascular status in acute hemorrhage, severe ulcerative colitis especially if complicated by toxic megacolon, myasthenia gravis, hiatal hernia associated with reflux esophagitis, hypersensitivity to any component of f he formulaf ion, acuf e infermiffenf porphyria. 

Compatible with previous findings (Perry et al, 2004 Perry and Soreq, 2004 Sklan et al, 2004), AChE-R appears to contribute to the maintenance of cholinergic homeostasis while accentuating neuronal inflammatory reactions. Because AChE-R accumulation reduces ACh levels, suppression of AChE-R overproduction can retrieve the blockade over pro-inflammatory cytokine production, suppressing the inflammatory status in treated subjects. 

Cicutoxin is concentrated in the roots but also may be found in aboveground parts. A mouthful of the root may be sufficient to kill an adult. Death results from status epilepticus, possibly caused by excessive stimulation of cholinergic receptors in the basal ganglia or brain stem.
