Initially, the neuroleptics were used to manage severe anxiety, agitation, and aggression in individuals with severe mental illness such as schizophrenia, a psychotic illness characterized by delusions, hallucinations, and disorganized, illogical thinking. The first neuroleptic used in schizophrenia was chlorpromazine (Thorazine) in 1952. Chlorpromazine was found to produce a tendency for sleep, but unlike the previous phenothiazines, it also produced a disinterest in patients regarding their surroundings and, with patients suffering psychiatric disorders, an ameliorative effect on the psychosis as well as a relief of anxiety and agitation. These observations suggested that chlorpromazine had potential for the treatment of psychiatric disorders. Thus, what started out as an attempt to improve antihistaminio activity ultimately resulted in an entirely new class of chemical entity useful in an unrelated disorder (27). [Pg.88]
