Chloroquine chemistry

Recently, it has been shown that chloroquine is a powerful inhibitor of the decarboxylation of ornithine (10.32) by this enzyme, a reaction that may play an important part in the therapeutic action (Konigk and Putfarken, 1983). The approach described below is different and promising it is based on modification via organometaUic chemistry of a known bioligand (tamoxifen, chloroquine) in such a way as to optimize its effeds. This selective targeting aims to increase efficacy via the organometaUic functionality, perhaps involving the formation of oxidized spedes proximate to the active site [66-73]. 

Recently, chloroquine has been successfully modified with a ferrocenyl entity incorporated widiin the side chain of the drug. This drug named ferroquine is active against chloroquine-resistant strains of die malaria parasite (38). This molecule is an example of the development of bioorganometallic chemistry in die design of drugs.
