The naturally occurring Z-butylidene phthalide also can be synthesised from the phthalaldehydic acid using Wittig reaction. 

The LD50 of intravenously administered ferulic acid in mice is reported to be 856.6 mg/kg (Ozaki and Ma 1990) of intraperitoneally administered ligustilide in mice is approximately 410 mg/kg (Xie and Tao 1985), and of orally administered 3-n-butylidene phthalide in rats is 2.45 g/kg (Opdyke 1979).

Lovage (L. officinale) accumulates in the leaves always a-terpinyl-acetate as main compound (40%-80%) besides a lower level of p-phellandrene (15%-28%). The main components of the roots 3-butylidene-phthalide and Z-ligustilide are only in lower concentrations—not rarely only in traces—present (Novak, 2006). The composition of the fruits is similar to that of the leaves with P-phellandrene as main component up to 60% (Bylaite et al., 1998).

The phytochemicals dihydrophthalic acid, ligusti-lide, butylidene, phthalide, and -valerophenone-0-carboxylic acid have been isolated from Angelica root (Angelica sinensis). They are likely to contribute to the observed circulatory modulating effects of Angelica root, including increasing coronary flow, modulation of myocardial muscular contraction, and antithrombotic effects.
