A DNA fragment, which may contain a specific gene, can be isolated from the cellular genome. Genes isolated from eukaryotic cells usually contain introns. 

The mRNA for a gene may be isolated, and a DNA copy (cDNA) produced. Single-stranded cDNA may be copied to form double-stranded cDNA 

The DNA that codes for a small protein may be chemically synthesized. 

A vector for transferring the foreign DNA into the bacterium, such as a plasmid, is cleaved with the same restriction endonuclease as the foreign DNA. The plasmid should contain genes for antibiotic resistance.
