Biomimetic pathway

Conjugated polyeneyne 136 is a model precursor for the intramolecular Diels-Alder cyclization in a biomimetic pathway to the esperamycins (Scheme 5-7). This model compound is synthesized via 133-135 by regiocontrolled sequential coupling of three different alkynes using two kinds of appropriate Pd catalyst [66]. 

Cephams have been prepared via a biomimetic pathway, i.e. the iron(III)-promoted cyclization of thiyl radical, as exemplified in Eq. (16) [35]. 

Attempts to synthesize nakadomarin A have focused on the stereoselective construction of the strained ABCD central core, which involves a reactive furan ring before accessing the EF rings (Scheme 20). We first investigated the synthesis of this strained 6/5/5Z5 tetracyclic core of nakadomarin A by two biomimetic pathways. We reported the first synthesis of the ABCD core of nakadomarin A in 2001, in which a novel intramolecular Mannich-type cycli-zation between a furan ring and an iminium cation was a key reaction [107]. If the molecule proves to be an effective ligand of interest, it will be s mthesized in the laboratory. The intellectual process to draw on the bios mthetic pathways is increasingly involved in the multistep synthesis of eomplex molecules. The produetion, at a laboratory but also an industrial seale, through biomimetic pathway helps to partially replace petrochemical procedures, based on the use of fossil resourees, by unique, inexpensive and often more seleetive procedures (synthesis of a given isomer or stereoisomer).

Mulzer s elegant synthesis of morphine is characterized by a stepwise elaboration of the ring motives in the natural product. While the biosynthetic pathway comprises an oxidative phenolic coupling, Mulzer employed a highly efficient Friedel-Crafts acylation to estabhsh the C12—C13 bond. Additionally, the implementation of observations from degradation studies represents an interesting alternative strategy to the exploitation of biomimetic pathways for natural product synthesis. 

Astonishingly, only eight nucleotide-activated donors are used by mammalian cells (see Scheme 2) [29]. In principal all are commercially available, although on a small scale only. The high prices of the natural donor substrates and the unavailability of non-natural derivatives of those donors is the reason why several chemical and biomimetic pathways have been probed to increase access to these crucial compounds [29, 30], The ensuing sections will summarize the most recent attempts to synthesize these donors, with emphasis on non-natural derivatives.
