The 4-[/V-(l-(4,4-dimethyl-2,6-dioxocyclohexylidene)-3-methylbutyl)amino]-benzyl (ODmab) esters display complete stability to 20% v/v piperidine in DMF and acidolysis (TFA), yet are readily removed with 2% v/v hydrazine monohydrate in DMF within minutes. However, it should be noted that this deprotection condition is sufficient to remove the A-Fmoc protecting group. Nevertheless, the ODmab ester has distinct advantages over OTmse and allyl esters, since its removal is facile (90, 91) and avoids extensive post-column manipulations.
