Antiglaucoma

A reaction of this type was used to prepare an intermediate in the synthesis of a natural compound with antiglaucoma activity.143... 

In the synthesis of Travoprost, an antiglaucoma agent, a bicyclo[2.2.1]heptan-2-one is converted to a lactone.240 The commercial process uses peroxyacetic acid as the oxidant and gives a 40% yield. The regioselectivity in this case is only 3 1 but the unwanted isomer can be removed by selective hydrolysis. 

Multifluorinated aromatics also play a significant role within bioactive compounds. Difluoroaromatics have already been discussed in Chapter 3. Bioactive tetrafluorobenzene derivatives are exemplified by the pyrethroid insecticides transfluthrin (6-9) and tefluthrin (6-10), whereas there are numerous pentafluorobenzene compounds that have proved of interest as potential insecticides, anticancer or antiglaucoma drugs (6-11,6-12, and 6-13) (Fig. 6.5).
