Anthrylmethyl esters are prepared from N-protected amino acids by reaction with 9-(chloromethyl)anthracene in acetonitrile and in the presence of TEA as base.t They are readily cleaved by acidic (2M HBr/AcOH, rt, 10-30 min) and alkaline (0.1 M NaOH/di-oxane, rt, 15 min) hydrolysis as well as by catalytic hydrogenolysis. These esters are very reactive toward thiolate anions (NaSMe, THE, HMPA, —20°C) with quantitative release of the carboxylate within 30 seconds to 1 hour.O l...
