Anthrone reagent

Anthrone reagent - dissolve 0.5 g anthrone in 500 ml sulphuric acid, 76% v/v. Prepare fresh daily and keep at 0-5°C. 

Because, on treatment with the anthrone reagents,224,225 hexoses and 5-(hydroxymethyl)-2-furaldehyde give solutions having identical spectral characteristics, dehydration is indicated to be the important reaction in this analysis. This conclusion is further supported by the reported isolation228 of 10-furfurylidene-9,10-dihydro-9-oxoanthra-cene (121) after reaction of 2-furaldehyde with anthrone, and by the fact that 121 has an absorption maximum of 600 nm, a value close to that used for pentose estimations. In similar studies,227 9,10-dihydro-10-(5-methylfurfurylidene)-9-oxoanthracene (122) was reported to have been isolated after the reaction of either L-rhamnose or 5-methyl-... 

At room temperature, add 4 ml of anthrone reagent, each to sample and standards. Vortex and let stand for 10 min, vortex again and leave for another 10 min before measuring the OD. 

The Dreywood100 anthrone reagent, which gives a positive qualitative test for a large variety of carbohydrates, as well as for furfural and its 5-hydroxymethyl derivative, gives a negative test for 2-desoxyribose.101...

Starch. The Anthrone reagent and iodine colorimetric methods described by McCready et al. (7) were employed to determine the starch and amylose contents respectively. A Perkin Elmer s Coleman 575 spectrometer was used to measure amylose at 660 nm, and starch at 630 nm.

The method is also readily adaptable to measuring carbohydrate in particulate material. It has not been employed in Section IV.8 since for phytoplankton the principal sugar has been shown to be glucose (Parsons, Stephens, and Strickland, J. Fish. Res. Bd. Canada, 18 1001, 1963), which gives the greatest colour with the anthrone reagent. 

Prepare exactly as described above for the anthrone reagent but omit the anthrone. The solution is stable indefinitely. 

Pipette 1.00 ml of the suspension, before any settling can take place, into a 20-ml Pyrex test tube. Add 10.0 ml of anthrone reagent from a pipette and mix the cells into the reagent with the buzzer or a clean glass rod. (Drain but do not rinse the rod.) Place the tube into a water bath or block heater at 100 C for a suitable period (Note b). 

The value of F should be approximately 2000 and should be determined at least once with each new batch of anthrone reagent.
