Amide bonds, cleavage, facile

In attempting a routine acid de-protection of a cyclic hexapeptide containing an iV-methyl-a -aminobutyryl residue, it was found that cleavage at this site of the peptide linkage also occurred. To study this facile amide bond cleavage, a model cyclic pentapeptide (231) was treated with neat trifiuoroacetic acid and it was found to suffer ready cleavage to the linear pentapeptide (232). The generality of this reaction and its mechanism was further explored by studies of a series of methyl... [Pg.108]

The phenol-sulfide SC linker 1.28 (85) attached to Merrifield resin has been used to support peptides, with the exclusion of S-containing aminoacids. Oxidation of the sulfur atom with hydrogen peroxide increases the reactivity of the linker towards amines and allows the facile cleavage of the ester bond to release the final compounds into solution as amides. [Pg.17]

The reaction of hexachlorodisilane with one or two equivalents of lithium bis(tri-methylsilyl)amide (LiN(TMS)2) affords the corresponding mono- or bisaminochlorodisilanes [4, 5]. Because of the stability of the Si-N bond towards LiAIH4 and its facile cleavage with dry HCl, amino-substituted silanes are valuable starting materials for partially hydrogenated chlorodisilanes. [Pg.346]
