Alpha2-adrenergic agonists

Alpha2 adrenergic agonists such as clonidine and guanfacine, which decrease sympathetic tone via central effects and feedback effects on norepinephrine release and which tend to produce an overall sedating effect, are used primarily to treat hyperaroused behavior, sleep disturbances, and tic-like behavior in autistic patients (Oswald and Sonenklar, 2007). Evidence for the efficacy of this class of drugs in autistic patients is limited.

In animal models, clonidine (alpha2 adrenergic agonist) has also been reported to prevent nerve agent-induced seizures (Buccafusco et al, 1988). Prophylactic use of clonidine may be limited by the marked ataxia and sedation produced by this drug in effective concentrations.
