Allylic alcohol synthesis from aldol reactions

In two studies toward the total synthesis of natural products it could be shown that the a,jS-unsaturated esters derived from the vinylogous Mukaiyama aldol reactions can be further functionalized into advanced intermediates. Asymmetric allylation of aldehyde 66, available in two steps from 3-hydroxypropionitrile, with allyl borane 67 gave the expected homoallylic alcohol 68 in good yield and with acceptable enantioselectivity. This material was converted uneventfully to a doubly protected dihydroxy aldehyde 69 and thence to 70 along lines similar to our synthesis of 56 from 34 (see Scheme 8). Since its discovery in 1974, more than 180 papers have been published on the use of the Overmann rearrangement to prepare allylic amines and their analogs from their allylic alcohols. For example, a recent synthesis of the fluoroalkene peptidomimetic precursor of A -acetyl-Z,-glutamyI-Z.-alanine involves the Overman rearrangement of the imidate derived from the allylic alcohol 60. ° This alcohol is readily prepared through Evans asymmetric aldol reaction of oxozolidinone with an aldehyde.
