Direct electrochemical determination of DNA is possible but without the differentiation of DNA specific sequences. Krznaric and Cosovic [166] were able to determine trout sperm DNA in the g/1 range by using AC voltammetry. They used a three-electrode system with a mercury drop electrode as a working electrode and platinum and calomel as the auxiliary and reference electrodes, respectively. They found a current peak at —1.4V for DNA. This peak was not significantly affected by RNA, dextran T-40 or bovine serum albumin which are commonly present either as contaminants or components of a hybridization solution. [Pg.400]
