In serial dilution experiment, Spiegelman [73, 74] and co-workers obtained three successive mutants with increased adaptation to presence of ethidium bromide, a drug that interferes with replication. Experiment starts with population of 10 MDV strands (variant of Q -RNA comprising about 220 nucleotides that is well-adapted to Q -RNA-replicase). Population is amplified to about 10 copies and subsequently diluted to initial concentration. Iteration of this procedure led to final product, three-error mutant that was obtained after about 40 iterations. As replication rate data show, mutant is slightly inferior to wild type in absence of ethidium bromide but twice as efficient as wild type at final concentration of ethidium bromide.

To determine the stability of the resistance phenotype, variant virus is serially passaged by limiting dilution, in the absence of drug. 

Measurement of serum CHE activity can serve as a sensitive indicator of the synthetic capacity of the liver. In the absence of genetic causes or known inhibitors, any decrease in CHE activity reflects impaired synthesis of the enzyme by the liver. A 30% to 50% CHE decrease is observed in acute hepatitis and in chronic hepatitis of long duration. Decreases of 50% to 70% occur in advanced cirrhosis and carcinoma with metastases to the liver. CHE is essentially normal in obstructive jaundice except when the cause is malignant. Serial measurement of CHE has been promoted as an indication of prognosis in patients with liver disease and for monitoring liver function after liver transplantation. 

Serial measurement of bilirubin is helpful in measuring the severity of liver disease. Bhirubiti fractionation is helpfiil only in jaundice of the newborn or in isolated elevations of bilirubin in the absence of other Uver test abnormalities that would indicate an inherited disorder of bilirubin metabolism. 

Figure 4 is the computer image of the 96-well microtiter plate. Each well is illustrated as a circle, and the thickness of the line used to draw the circle graphically displays the measured optical density in the wells. In the experiment illustrated, rows of weUs "A" and "H" were unused and are blmk. For each individual row, wells 3 through 10 illustrate a two-fold serial dilution of competitor. Wells in columns 1 and 2 for rows "B" through "G" are controls where the MelQx-albumin was omitted. The absence of color in these wells demonstrates the lack of non-specific binding of the anti-MelQx and peroxidase-anti-mouse antibodies, and defines experimentally "complete inhibition" for the competitive ELISA. Wells "Bll" through "G12" define the other extreme experimental condition, that of no inhibition. In these wells, no competitor was added and all of die anti-MelQx antibody was available to bind to the solid phase.
