TLR agonists induce cytokine production in the PTE module. The culture supernatants were collected after 24-h incubation, the cytokine levels in supernatants were analyzed by Q-plex cytokine array. The data shown in the figure are the relative cytokine production for each treatment after subtraction of the basal production in unstimulated PTE modules. Tltis is a shot of the underneath side of the mold we will be using for our demonstration. The bottom of the mold is recessed to provide air space for the vacuum. Notice the large and small holes drilled into the mold. These are vacuum draw holes that have been drilled through to the mold cavity. They are strategically placed and generally located at those places that require the deepe.st draw. There is a rubber seal glued around the outside bottom edge of the mold to prevent vacuum leaks between the mold and platen.
