Transfer Between Agar Deeps

Using a transfer loop, straighten out the loop, creating a straight wire. Hold the culture tube in left hand and transfer wire in right. Flame transfer wire, remove screw cap, and flame lip. 

Insert wire along channel created by previous stab, collecting a portion of the observed growth. Remove transfer wire and immediately flame lip and recap. Place back in rack. 

Remove the uninoculated deep from rack. Remove cap and flame lip. Immediately inoculate by carefully inserting the wire into the center of the agar plug and pushing the wire to the bottom of the tube. 

Remove the transfer wire. Flame test tube lip and recap. Flame sterilize transfer wire before proceeding. 

Where such culture techniques are routinely carried out, it is best to dedicate transfer wires exclusively for that purpose rather than straightening and reforming loops which shortens their useful period.
