Transaminase-Catalyzed Reactions with Whole Cells

5 Transaminase-Catalyzed Reactions with Whole Cells [Pg.733]

Reaction scheme for the synthesis of L-isoieucine with whoie ceiis. [Pg.734]

SCHEME 29.15 Reaction cascade for the synthesis of chirai amino aicohois in whoie ceiis. [Pg.734]

The reaction is driven by ammonium and glucose feed to the media. The NADH was regenerated by cellular metabolism from a carbon source like glucose. [Pg.734]

2-Keto-3-methyl-valerate (KMV) was the least inhibitory substrate up to 40 mM and was therefore chosen for the synthesis of L-isoleucine. The best reaction yields and almost complete conversion of 100 mM KMV were obtained with 50 mM MOPS, 100 mM (NH )2S0, and a glucose/KMV ratio of 1 2. In this reaction the overall yield of 0.6g/gh was obtained. [Pg.734]
