Culture tissue

Tissue cultures are useful for preliminary or specific work but are incomplete. 

In general, the specific constituents of milk are synthesized from small molecules absorbed from the blood. These precursors are absorbed across the basal membrane but very little is known about the mechanism by which they are transported across the membrane. Since the membrane is rich in lipids, and precursors are mostly polar with poor solubility in lipid, it is unlikely that the precursors enter the cell by simple diffusion. It is likely, in common with other tissues, that there are specialized carrier systems to transport small molecules across the membrane such carriers are probably proteins. 

In the appropriate culture medium, tissue explants give rise to callus tissue. Callus tissue is comprised of large, thin-walled parenchyma cells. It is similar to the undifferentiated tissue produced by plants as a repair mechanism when they are injured. In tissue culture, dedifferentiated callus can be induced to form plantlets that grow into normal plants. The induction of callus occurs when a sterile explant is brought into contact with a nutrient medium, which contains substances that initiate cell division and support growth. An explant may be a uniform piece of tissue or tissue derived from different cell types (Yeoman, 1973). Storage parenchyma tissue from Jerusalem 

Biology and Chemistry of Jerusalem Artichoke Helianthus tuberosus L. 

To create explants, dormant Jerusalem artichoke tubers are typically cut into 25-mm-thick slices, from which cylinders of tissue are fashioned. Cylindrical shapes are favored because they can be uniformly reproduced and have a high ratio of surface area to volume that optimizes gas and nutrient exchange, and facilitates callus formation. A small size is desirable to maximize the number of explants obtained from the same tissue source. Jerusalem artichoke tuber explant size is typically around 2.4 by 2.0 mm. The minimum size is usually 8 mg and approximately 20,000 cells (Yeoman, 1973), although some reports have used smaller explant sizes (e.g., Caplin, 1963). 

After the induction of tuber explants, cell division proceeds quickly. The cell number for a Jerusalem artichoke tuber explant increased 1000% over 7 days at 25°C (Yeoman et al., 1965). 

Electron microscopy (EM) has been used to study the development of Jerusalem artichoke explants in tissue culture. Tulett et al. (1969) described procedures for preparing tuber explants and callus tissue for EM to look at cell structure. Small pieces of callus were fixed in 6% glutaraldehyde in 0.1 M phosphate buffer at pH 6.9, at room temperature for 2 h, and then at 5°C overnight. After fixing, tissue was washed in several changes of phosphate buffer. Postfixation treatments involved immersion in a 1 to 2% buffered osmium solution for 1 h or a 2% aqueous solution of potassium permanganate for 1 to 2 h. 

The more challenging forms of tissue culture involve two or more types or animal cells grown simultaneously. Good progress has been made on skin replacements with products approved by the FDA Administration. Martin and Vermette (2005) discuss the reaction engineering aspects of tissue culture and conclude that the womb with its good mass transfer and essentially zero shear is ideal for growing complicated structures. 

40 Dogs weighing 20-30 kg were given 1 ml Combi-len i.m. 60 min before intravenous thiopental anaesthesia (5-lOmg/kg). After 1 mg succinyl cho-Une/kg intravenously the animals were intubated for nitrous oxide narcosis (NjO 02 =3 1). The anterior descending and circumflex branches of the left coronary artery were exposed and an Ameroid constrictor (Litvak et al. 1957) with 2.5 mm diameter was set. Epicardiectomy, cardio-pneumopexy, and omentopexy were performed to induce collateral circulation (for details of operation techniques see Wernitsch 1970).

Winkelmann (Borchen-Kirchborchen, Westphalia), Sprague-Dawley (Charles River, France) and Wistar rats of either sex were used in the experiments. They were fed Altromin R ad libitum. 

The recent development of culture systems for the growth and transformation of respiratory epithelial cells (Marchok etal. 1977, 1978 Terzaghi and Nettesheim 1979 Woodworth etal. 1982 Wu etal. 1982 Lechner etal. 1983 Pai etal. 1983 Thomassen et al. 1983 Nettesheim and Barrett 1984) will make it possible to study the potential transforming effects of silica particles directly on epithelial cells. It is possible that combined culture systems in which macrophages and/or cell cultures may provide versatile in vitro models to investigate cell to cell interactions and the role of silica in multicellular pathways. 

For the cultivation of airway epithelial cells, the cells are usually plated onto tissue culture plates coated with collagen or extracellular matrix components. Hormonally defined Ham s F12 medium contained 1 % penicillin-streptomycin, 5 ig/ml insulin, 5 ig/ml transferrin, 25 ng/ml epidermal growth factor, 15 ig/ml epithehal growth factor supplement, 2 X 10 M triiodothyronin, and 10 M hydrocortisone.

Although Brevundimonas (Pseudomonas) diminuta (ATCC 19146) is most commonly used for steriliziug-grade filter vaUdation, iu certain appHcations other bacteria are used. For example, when it is necessary to demonstrate removal of mycoplasma in appHcations involving sera and tissue culture media, membranes having a smaller pore size rating, eg, 0.1 p.m, are frequentiy used. For these membranes,laidlawii may be employed for vaHdation purposes (9). 

The plaque assay is desirable because it is very sensitive and only detects infectious viral particles. However, there are viral agents which cannot be supported by cell lines. In these cases other methods must be used. The polymerase chain reaction (PGR), which amplifies DNA or RNA from viral agents, can be used to detect the presence and quantity of viral agents. The amount of RNA or DNA target in the initial sample can be determined by competitive PGR where the quantity of amplified product is compared to a control PGR product where the initial amount of target is known. Quantification is also possible by an end-point dilution method similar to that used to determine a tissue culture infections dose. PGR methods can be very sensitive however. 

PVDE is a nontoxic resin and may be safely used in articles intended for repeated contact with food (190). Based on studies under controked conditions, including acute oral, systemic, subchronic, and subacute contact implantation and tissue culture tests, no adverse toxicological or biological response has been found in test animals (191,192). PVDE is acceptable for use in processing and storage areas in contact with meat or poultry products prepared under federal inspection and it complies with the 3-A sanitary standards for dairy equipment. 

In open fibers the fiber wall may be a permselective membrane, and uses include dialysis, ultrafiltration, reverse osmosis, Dorman exchange (dialysis), osmotic pumping, pervaporation, gaseous separation, and stream filtration. Alternatively, the fiber wall may act as a catalytic reactor and immobilization of catalyst and enzyme in the wall entity may occur. Loaded fibers are used as sorbents, and in ion exchange and controlled release. Special uses of hoUow fibers include tissue-culture growth, heat exchangers, and others. 

If the hGH is exported to the culture medium the product can easily be collected by removal of the cells from the culture medium by centrifiigation. Purification of hGH from the culture medium is faciUtated by low amounts of contaminating proteins present. In fact, it has been shown that hGH can be purified on a laboratory scale by a single purification step on a reversed-phase hplc column (43). Mammalian cells growing in tissue culture have also been used as hosts to produce hGH, which is exported into the culture media (44). 

In addition to the isolation of steroid raw materials from whole plants, plant tissue cultures have been iavestigated as an alternative source of these steroids. Despite many advances (128), there are no iadustrial appHcatioas of plant cell cultures for the production of steroids (129,130). 

Standardization and Testing". Potency is deterrained by titration of the amount of Hve vims Hi susceptible tissue culture and is mn Hi paraUel with a U.S. standard. Both in vivo and in vitro tests are used to assess safety (17). 

Aside from chemical methods, several patents have appeared on the biochemical production of natural vitamin from callus tissue cultures (41). 

Shikonin [517-89-5] (Cl 75535) occurs as an acetyl derivative in the Japanese shikone, Uthospermum eTythrorhi n another member of the Boraginaceae family. It is the (R)-optical isomer of alkannin (66). Tissue cultures of E. eythrorhi n are used in Japan to manufacture shikonin mainly for cosmetic use (67). Both alkannin and shikonin are mordant dyes producing violet to gray colors on fabrics. In Japan, shikonin was used to dye fabrics a color known as Tokyo Violet. Shikalkin [54952-43-1] the racemate (11), has been synthesized (68).

Commercial use of cell and tissue culture continues to expand. Improvement of organisms through recombinant nucleic acid techniques has become commonplace. Formerly, a few laboratories were well ahead of most others, but now the methods have been perfected for routine use. Another technique that is widely practiced is culturing of cells that excrete high concentrations of just one antibody protein. The specificity of antibodies and antigens is exploited in medical testing procedures using these pure monoclonal antibodies.

However, the complexities of plant cell and tissue cultures have led to revisions to this equation to include fresh cell weight and viability, cell expansion, and culture death phase. Semliki Forest arborvirus was grown in chick embryo tissue culture. The infectious tissue culture liquid was decanted and diluted with medium 199 to give a preparation containing between 10 and 10 mouse IDso virus/ml.

In an airlift fermenter, mixing is accomplished without any mechanical agitation. An airlift fermenter is used for tissue culture, because the tissues are shear sensitive and normal mixing is not possible. With the airlift, because the shear levels are significantly lower than in stirred vessels, it is suitable for tissue culture. The gas is sparged only up to the part of the vessel cross section called the riser. Gas is held up, fluid density decreases causing liquid in the riser to move upwards and the bubble-free liquid to circulate through the down-comer. The liquid circulates in airlift reactors as a result of the density difference between riser and down-comer. 

Rapid sterilisation of media is achieved by using microwave ovens. Most plant tissue culture media can be sterilised using a microwave, although it may not be suitable with a medium containing complex additives like oatmeal.
