Time Needed to Reach Equilibrium, Applied Voltage [Pg.42]

The small LKB column 8101 should not be allowed to dissipate more than 2-3 W at the beginning, the large LKB column, 6-9 W. The maximum permissible effect always diminishes as electrofocusing proceeds. This is discussed in Section VI, 10. [Pg.42]

Since the conductivity throughout the column diminishes as the carrier ampholytes reach their isoelectric points, the applied voltage can be increased considerably. Between 600-1000 V can be applied, depending on the type of pH-gradient used. [Pg.42]

The time required for electrofocusing depends on the pH range and the voltage used. A narrow pH range takes more time than a broad one. This is because the proteins have to migrate longer distances while carry- [Pg.42]
