Sulfite Oxidoreductase

There is some evidence that the iron-sulfur protein, FhuF, participates in the mobilization of iron from hydroxamate siderophores in E. coli (Muller et ah, 1998 Hantke, K. unpublished observations). However, a reductase activity of FhuF has not been demonstrated. Many siderophore-iron reductases have been shown to be active in vitro and some have been purified. The characterization of these reductases has revealed them to be flavin reductases which obtain the electrons for flavin reduction from NAD(P)H, and whose main functions are in areas other than reduction of ferric iron (e.g. flavin reductase Fre, sulfite reductase). To date, no specialized siderophore-iron reductases have been identified. It has been suggested that the reduced flavins from flavin oxidoreductases are the electron donors for ferric iron reduction (Fontecave et ah, 1994). Recently it has been shown, after a fruitless search for a reducing enzyme, that reduction of Co3+ in cobalamin is achieved by reduced flavin. Also in this case it was suggested that cobalamins and corrinoids are reduced in vivo by flavins which may be generated by the flavin... [Pg.106]

As with xanthine oxidase, the sulfido ligand of the active form of aldehyde oxidoreductase is readily replaced by an oxido ligand to yield a cofactor with a structure that resembles that of oxidized sulfite oxidase and assimilatoiy nitrate reductase. Both x-ray and EXAFS data are available for the bis(oxido) form, and, with the exception of the oxido replaced sulfido ligand, few changes are obvious in the overall structure of the oxidized form of the desulfo cofactor. Upon reduction of the enzyme the oxido ligand is presumably reduced to hydroxido, an observation that is supported by EPR data for the Mov state. [Pg.117]

The molybdenum-containing oxidoreductases that catalyze Eq. (1) have been variously termed molybdenum hydroxylases (6), oxotransferases (7), and oxo-type molybdenum enzymes (8). Molybdenum hydroxylase aptly describes the conversion of xanthine to uric acid, but the name seems less appropriate for the reactions catalyzed by sulfite oxidase and nitrate reductase oxotransferase implies that the function of these enzymes is to transfer oxo groups, even though relatively little is known about their actual mechanism of action and the name oxo-type molybdenum enzyme recognizes both the apparent oxo transfer chemistry of Eq. (1) and the fact that the molybdenum atom in each of these enzymes contains at least one terminal oxo group. In this chapter, we shall refer to these enzymes as pterin-containing molybdenum enzymes because a 6-substituted pterin appears to be a common chemical feature of all of the enzymes. [Pg.3]
