Sparfloxacin Antacids

Concurrent treatment with antacids reduces the oral absorption of many quinolones, such as ciprofloxacin and enoxacin (127), moxifloxacin (16), norfloxacin (128), ofloxacin (129), and sparfloxacin (130). 

The absorption of sparfloxacin is impaired by antacids containing aluminium or magnesium (19). In an open, single-dose (400 mg), randomized, four-way crossover study in 20 male volunteers (aged 18-38 years), Maalox (30 ml) given 4 hours after sparfloxacin did not cause a statistically significant reduction in the rate and extent of sparfloxacin absorption. In contrast, Maalox given 2 hours before or 2 hours after sparfloxacin did reduce its absorption AUC fell by 23 and 17% respectively and mean C ax by 29 and 13% (20).
