Separation of Nucleotides on Dowex Formate Columns

Prepare Dowex formate columns according to steps 4-1 to 4-8,4-16, and 4-17 to 4-23. [Pg.165]

Allow the column to drain and, when the level of the liquid reaches the top of the resin, add the mixture of the nucleotides (10 mg each of adenosine, AMP, ADP, and ATP in 1 ml solution adjusted to pH 7.9) to be separated. [Pg.165]

Repeat steps 4-29 to 4-48. This procedure 0-5N linear formic acid gradient) removes the AMP and ADP. [Pg.165]

Wash the column (using steps 4-65 to 4-66) using 5N formic acid containing 0.8N ammonium formate. This removes the ATP. [Pg.165]
