Purine ring mimics

To mimic the long bond length in the transition state, compound 12 was synthesized as a multisubstrate inhibitor (62). This compound mimics both substrates of PNP, the purine nucleoside, and phosphate. The two-carbon bridge between the ribose ring and the phosphonate group was incorporated to place the phosphonate moiety into the phosphate-binding site.
