Preparation of Agar Slants and Stabs

Prepare the suitable agar medium for the microorganism of choice. 

Heat the medium to dissolve agar (solution will clear indicating the agar has dissolved). 

While hot, distribute either 8mL (slants) or 10 mL (stabs) of medium into each 15 X 125 mm test tube. 

After removal from the autoclave, tighten the caps. 

For slants, lay tubes to allow the agar to solidify at an angle. Laying the cap-end on a meter stick works well.
