Potential Applications of the PROTAC Technique

Due to the fact that many diseases, including cancers, are dependent on the presence or over-expression of a small number of proteins, PROTACs designed to target these proteins could be of potential use in the clinic. Some of the most exciting applications of the PROTAC technique involve the exploitation of its many unique features in the development of PROTAC-based therapeutics. 

One exciting possibility is the development of therapeutic PROTAC molecules that would be active only in a particular tissue of interest. The identification of a ligand for an E3 ligase that is expressed in a tissue-specific manner would enable highly specific PROTAC-based therapeutics to be developed, minimizing potential off-target effects of the drug.
