Cells are grown in 10-cm culture dishes with 10% PCS to 40-50% confluency, and rinsed twice with Opti-MEM. For each culture dish, 4 fj,g of pEGFP plasmid are diluted with 30 /rl of Plus reagent, then mixed with 750 fil of Opti-MEM and incubated 15 min. In a separate vial 30 fil of Lipofectamine reagent are diluted in 750 /xl of Opti-MEM. Plasmid solution and lipofection mixture are then combined and allowed to stand for 15 min at room temperature (polystyrene tubes only). The plasmid/liposome preparation is finally poured into the culture dish and cells incubated at 37°C for 4 h, after which 3-5 ml of growth medium supplemented with 10% PCS is added to each dish, and incubation proceeds for 4 days. [Pg.198]
