Chan and Schellenberg (115) found evidence for its presence and reported its role in the enzymic reaction. Sequence analysis (39) has established that the enzyme is indeed devoid of tryptophan. Furthermore, Allen and Wolfe (116) have shown that SrMDH does not contain significant amounts of tritium after incubation with NAD-a 8 H or DL-[2- H]malate. Because of these findings, the report (117) that pig heart m-MDH is inactivated by 2-hydroxy-5-nitrobenzyI bromide, a reagent reported to be highly selective for tryptophan, must be attributed to modification at another site.
