One of the earliest diffraction studies was that of Wilkes et al. (1973) on neonatal-rat stratum comeum carried out using WAXD combined with differential thermal analysis. They also investigated the dependence of the diffraction pattern on the orientation of the sample. With the x-ray beam normal and perpendicular to the surface of the SC, two strong reflections, corresponding to repeat distances of 4.2 and 3.7 A, were observed that were assigned to crystalline lipids. In addition, two halos were observed, at 9.8 and 4.6 A, that were attributed to protein. Recently Al-Saiden et al [181] examined the thermal transitions in desiccated stratum comeum membranes of neonatal rats, rabbits and adult abdominal human skin. Four endothermic transitions were observed at 39-45°C, 55-58°C, 68-74°C and 77-86°C. The transitions at 39-45°, 68-74° and 77-86°C were attributed to phase charges in the intercellular lipid bilayers. A fifth transition at 48° was seen only in the neonatal rabbit stratum corneum and was attributed to lipid melting. [Pg.1008]
