Natural product chemistry

This case history presents only a simple account of one of R.B. Woodward s adventures based on ingenious undentanding of structural features and experimental findings described in the literature. The hydrogenation of porphyrins is still one of the most active subjects in heterocyclic natural products chemistry, and the interested reader may find some modem developments in the publications of A. Eschenmoser (C.Angst, 1980 J.E. Johansen, 1980). 

Synthetic utility of stereoselective alkylations in natural product chemistry is exemplified by the preparation of optically active 2-arylglycine esters (38). Chirally specific a-amino acids with methoxyaryl groups attached to the a-carbon were prepared by reaction of the dimethyl ether of a chiral bis-lactam derivative with methoxy arenes. Using SnCl as the Lewis acid, enantioselectivities ranging from 65 to 95% were obtained. 

Epoxide-opening iatramolecular cycli2ations, in light of their importance in natural product chemistry, have received much attention (62). 

The held of marine natural products chemistry, which encompasses the study of the chemical structures and biological activities of secondary metabolites produced by marine plants, animals, and microorganisms, began in earnest in the early 1960s. The underlying assumption driving marine natural products chemistry research is that secondary metabolites produced by marine plants, animals, and microorganisms will be substantially different from those found in traditional terrestrial sources simply because marine life forms are very different from terrestrial life forms and the habitats which they occupy present very different physiological and ecological challenges. The expectation is that marine organisms will utilize completely unique biosynthetic pathways or exploit unique variations on well established pathways. The marine natural products chemistry research conducted to date has provided many examples that support these expectations.

The natural world is one of eomplex mixtures petroleum may eontain 10 -10 eomponents, while it has been estimated that there are at least 150 000 different proteins in the human body. The separation methods necessary to cope with complexity of this kind are based on chromatography and electrophoresis, and it could be said that separation has been the science of the 20th century (1, 2). Indeed, separation science spans the century almost exactly. In the early 1900s, organic and natural product chemistry was dominated by synthesis and by structure determination by degradation, chemical reactions and elemental analysis distillation, liquid extraction, and especially crystallization were the separation methods available to organic chemists. 

The isolation and structural characterization of plant constituents are often difficult and time-consuming. Consequently, at the present time, only a small percentage of the total number of natural plant products has been identified. Early in the development of natural product chemistry it was apparent to many botanists and chemists that plants might be characterized and classified on the basis of their chemical constituents. The distribution of chemical constituents among plant families is presented by Swain (139) and Hegnauer (66). 

Note. In carbohydrate nomenclature, substitution at a heteroatom is normally indicated by citing the locant of the attached carbon atom, followed by a hyphen, and then the italicized heteroatom element symbol, e.g. 2-0-methyl, 5-N-acetyl. Substituents on the same kind of heteroatom are grouped (e.g. 2,3,4-tri-0-methy 1), and substituents of the same kind are cited in alphabetical order of heteroatoms (e.g. 5-N-acetyl-4,8,9-tri-0-acetyl). The alternative format with superscript numerical locants (e.g, N5,(/,(), ( -tetraacetyl), used in some other areas of natural product chemistry, is unusual in carbohydrate names.

Solving Problems with NMR Spectroscopy is a very welcome addition to the existing literature. It fulfills a real need for an up-to-date and authoritatively written introduction for students and practitioners of NMR. The vast experience of Professor Atta-ur-Rahman and Dr. Muhammad Iqbal Choudhary in the field of structural natural-product chemistry combined with a profound understanding of the concepts and techniques of NMR has led to a very useful and reliable treatise of practical NMR that is useful both for graduate students and for research workers. Professor Atta-ur-Rahman and Dr. Muhammad Iqbal Choudhary are congratulated for their admirable achievement. 

It is my opinion that this approach has considerable merit, provided that the questions posed in the problems are wisely selected, as indeed they are in this text. The authors themselves are well versed in natural-product chemistry, an area that presents a wide array of small molecule structural problems. They are therefore concerned that the reader reach the practical goal of applying the full power of NMR spectroscopy to problems of this type. To this end they have selected problems that address methods for solving structures as well as those that pertain to basic theory. The authors have wisely made a point of treating the more widely used ID and 2D experiments in considerable detail. Nevertheless, they also introduce the reader to many of the less common techniques.
