5- methoxy 4-methylenedioxyamphetamine

This was dissolved in 100 mL IPA, neutralized with con-centrated HCI (20 mL was needed) and diluted with 500 mL anhydrous Et20. More IPA was required to keep an oil phase from appearing. After the crystalline product was completely formed, it was removed by filtration, washed with IPA/Et20, and finally with Et20. Air drying gave 31.1 g of 2-methoxy-4,5-methylenedioxyamphetamine hydrochloride (MMDA-2) with a mp of 186-187 deg C. 

The residual white oil was dissolved in 250 mL anhydrous Et20, and saturated with anhydrous HCI gas. There was produced a crop of white microcrystals of 2-methoxy-3,4-methylenedioxyamphetamine hydrochloride (MMDA-3a) which was removed by filtration, washed with Et20, and air dried to a constant weight of 1.2 g. The mp was 154-155 deg C. 

The retention characteristics of sixteen phenylalkylamine derivatives (e.g., amphetamine, phentermine, 5-methoxy-3,4-methylenedioxyamphetamine, mescaline, methoxamine) were monitored on a 40°C C g column (photodiode array detector, A= 190-300nm) using a complex 22-min 5.5/94.5-> 39/61 (90/10 acetonitrile/water [5mL/L H3PO4 and 0.28 mL/L hexylamine])/water (5mL/L H3PO4 and 0.28 mL/L hexylamine) gradient [1425]. Urine and cactus samples were extracted and detection limits of 25-50 ng/mL and 40ng/g, respectively, were reported.
