Methods recommendations

The spectrophotometric method recommends using standard solutions of phosphate in the range of 2-10 ppm, whereas the FIA method recommends standards in the range of 10-60 ppm. Explain why the methods use a different range of standards. 

If the mutual solubilities of the solvents A and B are small, and the systems are dilute in C, the ratio ni can be estimated from the activity coefficients at infinite dilution. The infinite dilution activity coefficients of many organic systems have been correlated in terms of stmctural contributions (24), a method recommended by others (5). In the more general case of nondilute systems where there is significant mutual solubiUty between the two solvents, regular solution theory must be appHed. Another potential problem with cyanide analysis is the recommended preservation method. The APHA standard method recommends preservation by adjusting to a pH of 12 using sodium hydroxide. The Department s laboratory has been using this method which is effective for total cyanide but unsatisfactory for free cyanide since the pH adjustment can change the cyanide species present, and thus the final result. There is no adequate preservation method for free cyanide. 

The methods recommended above for mitigation of on-farm GHG emissions in the fluid milk supply chain were obtained from years of research that evaluated the impact of various practices on each GHG. The research was typically conducted on experimental farm plots under a particular farm management system meaning that the data are relevant for that system with a particular soil type, herd size, and climate, for example, and may not apply to a different farm management system. The impact of a particular mitigation procedure for one GHG on another is also relatively unknovm as well as the impact of the mitigation on more complex interactions such as the total C and N throughout the farm. 

A sample of alfalfa hay from Bozeman, Mont., that had received two spray applications, each of 4 pounds of technical toxaphene per acre, was found by the method recommended by Carter and Hubanks (3) to contain 225 p.p.m. of organic chlorine, equivalent to 331 p.p.m. of toxaphene. This sample was obtained from a bale approximately 5 months after the last spray application. A similar sample of untreated hay was found to contain 1.2 p.p.m. of organic chlorine. 

Samples of abdominal fat from a steer that had been fed hay containing toxaphene residues and from a steer that had been fed untreated hay were analyzed for their organic-chlorine content by the method recommended by Carter (i). The organic-chlorine content of the fat from the treated steer in excess of the amount found in the untreated steer was equivalent to approximately 700 p.p.m. of toxaphene. 

The following protocol is based on the methods recommended by Thermo Fisher for use of the cyanine dye DyLight 649. Antibody reduction is based on the methods of Sun et al.
