Many scientists would argue that the flexibility and reliability of SPA more than offset these potential disadvantages, and SPA is still in wide use for HTS today. In the absence of cAMP the holoenzyme complex is intact and C and R subunits, captured to AlphaScreen donor and acceptor beads, are brought into proximity generating a luminescence signal. IfcAMP or a cAMP agonist is added, the signal decreases as the subunits and thus the beads, dissociate (Figure 8.4B). 

Silicon oxide nanostruaures are prepared by local anodic oxidation on dodecyl-terminated silicon. Signal generation in almost all immunoassays requires attachment of a label to one of the reagents. This label can be (a) a radioisotope such as or (b) an enzyme, for example, horseradish peroxidase or alkaline phosphatase, (c) a fluo-rophore such as fluorescein, (d) a luminescent species such as luminol or (e) micro-or nanoparticles made of latex or gold.
