Hong et al. (2004) also found that modification of PAMAM dendrimers with a short PEG linker arm could act to reduce nonspecificity caused by the amines on the dendrimer-modified surface. An azido-PEGj-aininc spacer was activated with nitrophenyl carbamate to yield an activated intermediate that could be used to modify the amines on the dendrimer (Figure 7.24). Reaction at high molar ratio resulted in about 61 PEG-azido spacers on the dendrimer. Elimination of HI, which in the presence of an excess of IN3 can form hydrazoic acid, followed by its addition to the vinyl azides can give an intermediate triazide 75. The same compound could arise directly by substitution of one iodine atom by an azido group. The intermediate 75 has been considered to undergo a transannular reaction with homolytic cleavage of the weak C—I bond to form the radical 76, which loses a nitrogen atom to a radical 77. Combination of the two radicals leads to the 2-tetrazene 74 (equation 77). 

The radical adduct obtained by the addition of (TMS)3Si to the keto azide 66 underwent an opening of the cyclopropyl ring, with formation of 67 as intermediate, prior to the intramolecular addition to the azido group [60]. After tosylation the final product was obtained in 61 % yield. 

Starting from acylated 2-azidomethylbenzimidazoles (145), an additional imidazole ring can be condensed by transformation of the azido group with tri-n-butylphosphane into the appropriate iminophosphorane intermediate 146. After extrusion of phosphane oxide, cyclization occurs to the 1-substituted 4//-imidazo[l,5-a]benzimidazole 147 (Scheme 58) (89T1823 94S1197). 

An example of ring enlargement is the intramolecular Schmidt reaction of azidoalkyl ketone 53 that gives, by the action of titanium tetrachloride, through intermediate azidohydrin, lactam 20 (95JA10449). Because of the large distance between keto and azido groups, the usual catalyst trifluoroacetic acid does not work. 

Based on results presented in Scheme 37, Logothetis suggested that the thermal decomposition products from the olefinic azides in the scheme are derived from triazoline intermediates formed by an intramolecular cycloaddition reaction and not by fragmentation of the azido group to a nitrene.100 However, allyl azide and 4-azido-l-pentene do not undergo internal cycloaddition because of the strain in the corresponding triazoline they fail to give aziridines and imines upon thermolysis.100... 

Labile annelated 5-aminotriazolines are also postulated as intermediates in the intramolecular reaction of the azido group with the 5,6 double bond of pyrimidines (Scheme 57).235,236...
