Ineffectiveness, therapeutic

There are innumerable reasons why the administration of a drag can lead to unexpected occurrences, some of which may be harmful. For example, hypersensitivity reactions may occur. These reactions may only show up in 1 in 10,000, meaning that they would be probabilistically unlikely to have been seen in pre-approval clinical trials, and they may show a remarkable absence of a relationship between dose and severity (Olsson and Meyboom, 2006). The underlying mechanism for this reaction may be an inborn variation in metabolism, but in many cases the etiology is unknown. Another example is therapeutic ineffectiveness. Therapeutic ineffectiveness is not often considered to be a side-effect, but it is one of the most common unintended responses to a drag. It is a recognized reportable event in pharmacovigilance, especially when it occurs unexpectedly (Olsson and Meyboom, 2006). [Pg.204]

GRAPEFRUIT JUICE BETA-BLOCKERS -CEUPROLOL (not available in USA) Very likely to be ineffective therapeutically due to a great i plasma concentrations (AUC i by 85% and maximum concentration by 95%) Attributed mainly to marked L absorption. This may be due to physicochemical factors or due to an inhibition of drug uptake transporters Do not co administer... [Pg.729]
