Environmental and human-safety aspects of surfactants have begun to receive considerable attention (19—21). 

Toxicity studies on trifluoroethanol show acute oral LD q, 240 mg/kg acute dermal LD q, 1680 mg/kg and acute inhalation L(ct) Q, 4600 ppmh. Long-term subchronic inhalation exposure to 50—150 ppm of the alcohol has caused testicular depression in male rats, but no effects were noted at the 10 ppm level (32). Although the significance of the latter observations for human safety is unknown, it is recommended that continuous exposure to greater than 5 ppm or skin contact with it be avoided.

NOAEL (no-observed-adverse-effect level) is defined as the highest dose at which no adverse effects are observed in the most susceptible animal species. Acceptable Risk. Once information is assembled concerning the characteristics of exposure and biologic effects, that information must be interpreted in terms of human safety standards. That interpretation requires that one establish a set of criteria representing acceptably safe conditions for human existence, bearing in mind that zero concentrations of environmental chemicals are unrealistic. 

Pesticides, including insecticides, herbicides, and fungicides, are widely used in agriculture, and the potential for these residues to accumulate in food has led to concern for human safety. Pesticide residues may enter food animals from environmental sources or from treated or contaminated feeds. Immunoassay development for pesticides has had major impacts for pesticide registrations, analysis of residues in foods, monitoring environmental contamination, determination of occupational exposure, and integration of pest management. 

Other agents that appear to be safe for use in pregnancy include the proton pump inhibitors, sucralfate, and meto-clopromide (Table 44-5). The proton pump inhibitor with the largest body of human safety data during pregnancy is omeprazole.24... 

Although these are all used across Europe, the hierarchy of these has altered slightly due to the influences of environmental legislation. For example, Chloro-isothiazolone has been identified as a skin sensitiser and hence therefore lost its number one position as a controlling agent. New actives such as the Octyl - isothiazolone have recently been introduced as a more stable molecule where it is hoped that lower levels can be used reducing environmental and human safety aspects. 

Materials that are under the OTC classification must be reviewed by the appropriate FDA division to evaluate elements concerning labeling, quantities of active ingredient, animal safety data, human safety data, and other elements that would support the marketing of the appropriate products. 

It was usual in the USSR that, even when strictly complying with the MPC, neither environmental nor human safety were ensured. Table 1.6 shows MPC examples of the water from reservoirs where the Health and Epidemiological Service needed to toughen MPCs in later years because several factors had been neglected. Shaded pesticides in this table (as well as following tables) were subsequently officially banned.

For many substances the body s own mechanisms for de-toxification and repair mean that low doses of some chemicals can be tolerated without experiencing any adverse effects. However, once a certain threshold has been exceeded then the degree of adverse effect is related to the dose. The highest dose at which no adverse effects are observed in the most susceptible animal species is identified as the No Observed Adverse Effect Level (NOAEL). Carcinogenicity studies are a time-consuming (up to 3 years to complete) and expensive (can be in excess of 1 million dollars each) proposition and are typically carried out in rats and mice. Their purpose is to determine if the drug possesses the capability to initiate or promote the development of tumors. The application of the results of these studies to the human safety has been debated for many years. In many instances over the past two decades, mechanistic studies have shown that positive responses in these rodent models do not have specific relevance for humans, and drugs have been approved on the basis of these explanations. While the scientific debate about relevance of these studies continues, they remain required by regulations. Positive responses without adequate explanation or safety margin can result in nonapproval of the product. 

This exposure relationship is frequently more important in establishing human safety margins, as dose alone may be subject to a variety of differences between species such as absolute bioavailability, distribution, and excretion. This aspect, now commonly referred to as "toxicokinetics," has been outlined in an ICH guideline.6 This guideline specifies minimum requirements in terms of number of time points examined, number of animals per time point, and the requirements for calculation of various pharmacokinetic parameters such as Cmax, AUC. These will become important for comparison with human data as it becomes available later.
