How to Produce Gradients Covering a Narrow pH Range

At present, the narrowest range commercially available covers 2 pH units. The manufacturer (LKB-Produkter AB, S-161 25 Bromma, Sweden) states that narrower pH ranges will soon be available. However, even when gradients covering a single pH unit can be made with the help of a commercial package, it will sometimes pay to use a still narrower [Pg.45]

In Method A the sample is present during two electrofocusing runs. The narrow range will be chosen depending on the relative position of the sample in the first run. [Pg.46]

In Method B a predetermined narrower range is taken out of a pre-run without the sample, and the sample is introduced in a second gradient made up by using the selected portion of the first run. [Pg.46]

When conditions permit, Method A is preferable since the exact location of the interesting sample is used as a means of selecting the range. Uninteresting components are eliminated in the first run. [Pg.46]

To obtain these special narrow-range pH-gradients, it is recommended that the range 6-8 be added in an amotmt corresponding to one tenth ( ) of the total amount of carrier ampholytes. [Pg.46]
