There are two principal modes by which drugs can bind non-covalently to DNA intercalation and external groove binding. 

Non-intercalative groove binding. The drug lies in one of the grooves in DNA, following the helical contour of DNA as it binds. Netropsin and dlstamycin bind in this way.
