Glycine-rich loop

The authors expressed PKA consisting of 353 amino acids, of which eight are prolines. Resonances of 274 backbone amide peaks were visible in the spectrum, of which 191 were assigned. It was possible to assign resonances for the N- and C-terminal sequences, the majority of the N-lobe, including the glycine-rich loop, and most of the solvent-exposed residues of the C-lobe. This enabled a determination of the structure for the more flexible parts of the structure. However, many correlations were missing for the... [Pg.25]

The catalytic cores of protein kinases present in eukaryotic beings share the same fundamental structure, best defined for cAMP-dependent protein kinase (PKA) and characterised by an amino terminal 3-sheet-rich nucleotide-binding domain and a larger, helical peptide-binding domain. In the nucleotide-binding domain there is a flexible glycine-rich loop, absent in prokaryote kinases, that wraps the nucleotide and whose phosphate-... [Pg.821]

Figure 2-4 Informational content analysis of the ALAS2 glycine-rich loop. Functionally acceptable amino acid substitutions at each position are indicated. The wild-type murine ALAS2 glycine-rich sequence is shown in a shaded box. Results obtained with random mutagenesis and glydne-rich loop codon specific libraries are indicated above and below the wild-type sequence, respectively. (Adapted from [86]).
