Choline kinase phosphorylates choline to give a phosphocholine and participates in glycine, serine and threonine metabolism and glycerophospholipid metabolism. Hemicholinium-7 is the prototypical tool compound used to inhibit CHK. Based on inhibitor studies, it has been proposed that CHK is important for the regulation of cell proliferation. Inhibition of choline kinase is also used to target plasmodium and develop novel antimalarials. A series of papers on pyridinium based inhibitors have been published, but no disclosures of more drug-like molecules have been made. [Pg.195]

A role for DAG in tumorigenesis is supported by molecular studies that have correlated overexpression of the small GTPase oncogenes ras or sis with an elevated DAG level and the transformed phenotype (Preiss et al, 1986 Fleischman et al, 1986 Wolfman and Macara, 1987). The increase in DAG concentration was concluded to derive from an enhanced rate of phosphohpid turnover and not from the de novo synthesis of glycerophospholipids (Fleischman et al., 1986). These data implicate the abnormal activity of glycerophospholipid metabolizing enzymes in the progression towards the transformed state. [Pg.51]
