In the literature of fluorescent proteins the terms fluorescence and brightness are often used as synonyms to describe the detected light emission on protein or cell level. Since the usage of these terms is not consistent literature sometimes is misleading especially when it... 

Thorough photophysical FCS analyses have been carried out on various fluorescent proteins so far EGFP, YFPs, DsRed. These studies produced valuable information for fluorophore usage as fusion partners and successive data evaluation. They also document which photophysical parameters of reporter molecules ought to be determined in vitro to reduce the number of unknown parameters in vivo. We briefly outline these fluorophore studies and continue to hint to impUed options and limitations. 

Two photon excitation (TPE) and its application to PCS analysis has been described and discussed in detail and shall be briefly reviewed with regard to fluorescent proteins and intracellular usage [6,7,86,87].
